Sanfilippo sendromu olarak da bilinen MPS III, heparan N-sülfataz, α-N-asetilglukozaminidaz, asetil CoA:α-glukozaminid N-asetiltransferaz ve N-asetilglukozamin-6-sülfataz adlı 4 enzimin 1'ininin eksikliğinden kaynaklanmakta olup, bunlar hastalığın sırasıyla MPS IIIA, IIIB, IIIC ve IIID adlı 4 alt tipiyle ilişkilidir. Sonuç olarak glikozaminoglikan (GAG) heparan sülfatın hücre içinde birikimi, merkezi sinir sistemi dejenerasyonunun belirgin şekilde görüldüğü, progresif, multisistemik hastalığa neden olur.1,2
